The first batch of hundreds of migrants saved from the Mediterranean Sea arrived in Spain a few days ago and Italy and Malta refused to welcome them.
Hundreds of police, health officials and interpreters have been deployed to help receive migrants.
Following the rescue of migrants last week, there was a dispute among European countries over who should receive them.
Three ships carrying 629 migrants rescued by the ship Aquarius near the Libyan coast are due to reach the Spanish coast.
The new Socialist Spanish government promised to provide free medical care to immigrants and to investigate each asylum application separately.
Earlier this week, Prime Minister Pedro Sanchez said, "It is our duty to help prevent a humanitarian disaster and provide a safe haven for these migrants in compliance with our human rights obligations."
Sanchez has taken a less hawkish stance on the issue of immigrants since he took office two weeks ago.
Approximately 1,000 Red Cross staff will assist in receiving migrants on the wharf of Valencia.
Among the rescued migrants are 123 unaccompanied minors, 11 children under the age of 13 and seven pregnant women, officials say.
